Effect of supplementation with resveratrol on biomarkers associated with atherosclerosis in humans (2020)

  • Abstract: Background: Several current trials have investigated the effect of resveratrol supplementation on cardiovascular-related biomarkers. However, there is no consensus regarding its efficacy in humans, mainly due to several factors including dosage, intervention time, and studied population. Our objective was to conduct a review applying a multivariate statistical approach to identify the experimental conditions associated with the results presented in studies in which humans received resveratrol supplementation. Material and methods: The literature search included Pubmed, Science Direct, and Google Scholar databases. It was included the studies published in the last 10 years that investigated the impact of resveratrol on atherosclerosis-related biomarkers. Biochemical data were collected from 27 studies on the baseline and after the intervention time. From 81 biomarkers evaluated in at least one study, 12 biomarkers were selected for the multivariate analysis. The net change of these 12 biomarkers was calculated. Principal component analysis (PCA) followed by cluster analysis was applied to visualize the association among the biomarkers and also for assays grouping. One-way ANOVA followed by Tukey test was applied to compare the protocol parameters and biomarker's net change among the groups. Results: Resveratrol reduced total cholesterol, LDL cholesterol, triglycerides, and blood pressure, while increased HDL cholesterol. It was also identified that resveratrol reduced high-sensitive C-reactive protein (hsCRP) and tumor necrosis factor-α (TNF-α), but not interleukin (IL) 6 or leptin. In contrast, resveratrol improved the concentration of IL-10 and adiponectin, which are involved in anti-inflammatory responses. Applying multivariate analysis, we separated the studies into three clusters. The groups showed that the resveratroleffect could be dependent on protocol parameters, such as health conditions of the patients and intervention time. Conclusions: In general, the supplementation with resveratrol improved the lipoproteins profile, arterial pressure and inflammatory biomarkers. Regarding to the study design, our analysis showed that a longer intervention time can be necessary to achieve HDL increase in the patients
